Une fois activs, ces rcepteurs signalent depuis la membrane jusqu'au noyau par l'intermdiaire de mdiateurs cytoplasmiques appels Smads. La voie de signalisation de l'acide rtinoque (AR), elle, implique une internalisation du ligand, lequel est achemin jusqu'au noyau o il se lie des rcepteurs nuclaires, les RARs. Les effets de l'AR et du TGFbeta sur la croissance ou la diffrenciation cellulaire se rvlent synergiques ou antagonistes selon le type cellulaire. Ceci suggre que les voies de signalisation de l'AR et du TGFbetasont troitement lies. Ces travaux de thse montrent une interaction fonctionnelle entre les voies de signalisation de l'AR et du TGFbeta travers leurs mdiateurs, RARgamma et Smad3. Sur le plan physiopathologique, cette tude indique que les agonistes des RARs exercent une activit antiTGFbeta alors que les antagonistes potentialisent son action. Ceci suggre la possibilit d'une utilisation thrapeutique nouvelle de ces ligands des RARs dans des pathologies impliquant le TGFbeta telles que la cicatrisation anormale ou la fibrose
