The present investigation was under taken to evolve an efficient method for Indica rice transformation. Agrobacteriummediated methods used for rice transformation .Embryogenic calli were produced from mature seeds of IR64 and swarna (Indica varities) were used as a target cell in transformation experiments. MS medium supplemented with 2,4D, (2mg/l) kinetin, (0.4mg/l). L. Proline, (500mg/L) Casein Hydrolysate, (300mg/L) agaragar (0.8%w/v) was found best callus medium for swarna. MS medium supplemented with 2,4D, (2mg/L) LProline (500mg/L) maltose (30g/L) agaragar (0.8%w/v) was found to be best callus medium for IR64. Embryogenic calli were co cultivated with the agrobacterium strain LBA (4404) EHA105 using the protocol developed by Hiei et al, (1994) which was tested for applicability to IR64 and swarna Transformation.GUS assay was done with transformed calli and was better in swarna.
