Reversible Proteinphosphorylierung ist ein Schlsselprinzip der intrazellulren Signaltransduktion. OnesourcePeptid/Phosphopeptidstandards wurden mit dem Ziel entwickelt, Phosphorylierungsgrade von Proteinen positionsspezifisch und mit hoher Genauigkeit zu bestimmen. Kernpunkt der Methode ist die Erzeugung einer Mischung eines stabilisotopenmarkierten Peptid/PhosphopeptidStandardpaars, dessen stchiometrisches Verhltnis exakt bekannt ist. Die Standards wurden Proteinverdaus zugesetzt und diese mittels Flssigchromatographie und Massenspektrometrie analysiert. Im Vordergrund dieser Arbeit steht die Analyse der dualen ERK1/2Phosphorylierung durch die MAPKKinase MEK. Die ERKSignalkaskade spielt eine zentrale Rolle in der Antwort von Zellen auf extrazellulre Stimuli wie Wachstumsfaktoren und Zytokine. Dennoch sind die Mechanismen der dynamischen ERKAktivierung und Deaktivierung in vivo nicht vollstndig verstanden. Mit Hilfe von onesourceStandards wurde erstmals ein zweistufiger, distributiver ERKPhosphorylierungsmechanismus in primren Maushepatozyten nachgewiesen.
