Atorvastatin gehrt zu einer Gruppe lipidsenkender Medikamente, die ber eine Hemmung der HMGCoAReduktase wirken und daher zur Therapie der Hypercholesterinmie und der koronaren Herzkrankheit indiziert sind. In zahlreichen klinischen Studien konnte gezeigt werden, dass unter Statinmedikation ebenfalls die Schlaganfallinzidenz gesenkt wird und durch Statinvorbehandlung die klinischen Auswirkungen des Schlaganfalls gemildert werden. Dies ist umso erstaunlicher, als ein hoher Cholesterinspiegel als Risikofaktor fr den ischmischen Schlaganfall gar nicht als etabliert anzusehen ist, sondern kontrovers diskutiert wird. In zahlreichen experimentellen Studien zeigt sich aber, dass die durch Statine vermittelten Wirkungen vielfach auf cholesterinunabhngigen Mechanismen beruhen. Diese sogenannten pleiotropen Effekte umfassen vasodilatative, antiinflammatorische, antithrombotische und antioxidative Mechanismen. Gegenstand dieser Arbeit ist es, unabhngig von vaskulr vermittelten Komponenten die direkte Neuroprotektion eines Statins in neuronalen Zellkulturen zu untersuchen.
