Die Bedeutung der ?Arrestine als multifunktionelle Adapterproteine GPCRvermittelter Signaltransduktion hat in den letzten Jahren immer mehr zugenommen. In der vorliegenden Arbeit lag der Schwerpunkt auf der Untersuchung der molekularen Basis sowie der Ligandenabhngigkeit sowohl der ?Arrestin/RezeptorInteraktion als auch ?Arrestin(un)abhngiger Signaltransduktionsmechanismen. Durch Mutationsanalysen konnten Ser 352/Thr 358 im distalen CTerminus des P2Y1R als Schlsselstellen der ?ArrestinTranslokation und Internalisierung identifiziert werden, whrend ein oder mehrere Phosphorylierungsstellen im proximalen P2Y1R CTerminus die molekulare Grundlage der Rezeptordesensibilisierung darstellen. Im Gegensatz dazu stellen die Phosphorylierungsstellen zwischen den Positionen 355 und 364 essentielle Bereiche der ?ArrestinTranslokation an den ?2AR dar. Darberhinaus wurde gezeigt, dass die endogenen Agonisten am ?2AR bzw. dem P2Y2R verschiedene mit dem jeweiligen Rezeptor verknpfte GProtein bzw. ?Arrestinassoziierte Signalwege zu unterschiedlichem Ausma aktivieren (?biased agonism?).
