Das Serumglykoprotein Alpha 1Antitrypsin (A1AT) hat durch seine inhibitorische Aktivitt gegenber der Neutrophilen Elastase eine besondere physiologische Bedeutung in der Lunge. Bei der A1ATMangelErkrankung kommt es bei Patienten zu einer gesundheitsgefhrdenden niedrigen Konzentration des SerinproteaseInhibitors und einer erhhten Gefahr, an einem Lungenemphysem zu erkranken. Zurzeit werden Patienten mit A1AT behandelt, welches aus humanem Plasma isoliert wird. Neben der Gefahr einer Infektion mssen Patienten aufgrund der schnellen Degradation von A1AT wchentlich behandelt werden, um ein schtzendes Level an A1AT im Serum aufrecht zu erhalten. Eine bedeutende postranslationale Modifikation fr die Serumhalblebenszeit wird in der Glykosylierung von Serumglykoproteinen gesehen, da Asialoglykoproteine vom Asialoglykoproteinrezeptor in der Leber erkannt und aus der Zirkulation entfernt werden. In dieser Arbeit wurde der Einfluss von zustzlichen NGlykosylierungsmotiven, nicht natrlichen Monosacchariden und verschiedenen Expressionssystemen auf die Serumhalblebenszeit und Bioaktivitt von A1AT untersucht und eine Verbesserung der Serumhalblebenszeit von A1AT besttigt.
