Fr die Entwicklung eines plastidren Induktionssystems wurden bakterielle Elemente des quorum sensing in Nicotiana tabacum (Tabak) getestet. Die regulativen Komponenten wurden von Vibrio fischeri entnommen und an die Expressionsmaschinerie der Plastiden adaptiert. Mit einem hochsensitiven Glucuronidase (GUS)Assay wurde die Expressionsstrke des Induktionssystems in den transgenen Pflanzen ermittelt. Im nichtinduzierten Zustand konnte die Expression durch die Auswahl geeigneter Promotorelemente auf eine Konzentration von 0,0003% vom gesamtlslichen Protein erheblich gesenkt werden. Eine signifikante Steigerung der GUSExpression mit dem spezifischen Induktor wurde nicht erzielt. Mgliche Ursachen dafr werden anhand der Literatur diskutiert.Weiterhin wurde ein transientes Expressionssystem etabliert. Mittels Polyethylenglykol (PEG) wurden TabakProtoplasten mit verschiedenen GUSExpressionsvektoren transformiert und reproduzierbar eine signifikante GUSAktivitt gemessen. Probleme bei der Analyse von PlastidenExpressionsvektoren werden aufgezeigt und Lsungsanstze diskutiert.
