Das Auftreten von disseminierten Tumorzellen (DTCs), unter denen die Metastasenvorluferzellen vermutet werden, geht beim sophaguskarzinom mit einer schlechten Prognose einher. Die Untersuchung von solchen sophagealen DTCs knnte daher zur Identifizierung von therapeutisch relevanten Zielstrukturen beitragen. Zunchst wurde anhand einer Doppelimmunfluoreszenzfrbung die Verteilung von CK und EpCAM in/auf DTCs aus Knochenmark (KM) und Lymphknoten (LK) untersucht. Dabei konnte nachgewiesen werden, dass bereits publizierte genomische Unterschiede zwischen CK+DTCs aus KM und EpCAM+DTCs aus LK mit dem Detektionskompartiment und nicht mit dem Detektionsmarker einhergehen. Des Weiteren konnten metaphasebasierte CGHUntersuchungen das hufige Auftreten von Amplifikationen auf Chromosom 17, vor allem bei den prognostisch relevanten EpCAM+DTCs, besttigen. Um mgliche therapeutisch relevante Gene auf Chromosom 17 weiter einzugrenzen, wurde eine hochauflsende BACaCGH fr einzelne DTCs etabliert, optimiert und validiert. Die etablierte aCGH ermglichte es u. a. DTCs anhand der prognostisch relevanten Zielstruktur HER2 in HER2amplifizierte und HER2normale DTCs zu klassifizieren.
