Zusammenfassung In der vorliegenden Arbeit wird der Mechanismus der Entstehung der neutrophil en Granulocyten behandelt und eine neue Theorie tiber die Pro liferation der Granulocytopoese entwickelt. Bei dem typischen Wechsel gewebe, das verschiedene Reifungsstadien durchlauft, setzt sich im Gegen satz zu vielen anderen Geweben die Proliferation aus Regeneration und Differenzierung zusammen. Die Beurteilung der Proliferationsaktivitat aus dem Knochenmark wird in vivo weiterhin durch die nicht tiberschaubare GroBe der Ausschwemmung erschwert. Daten tiber die morphologischen Veranderungen und Zeitverhaltnisse bei der Entwicklung der neutrophilen Granulocyten werden zusammenge stellt und durch unsere experimentellen Untersuchungen an in vitro tiber lebenden menschlichen Knochenmarkzellen erganzt. Durch direkte Beobach tung mit dem Phasenkontrastmikroskop wurden Ergebnisse tiber die Dauer der Mitosen, der Interkinesen und der Reifung vom Promyelocyten zum Segmentkernigen gewonnen. Die reifende Granulopoese wies bei einem in vitro beobachteten Fall eine Reifungszeit vom Promyelocyten zum Segmentkernigen von 4 Tagen und eine Generationszeit von 30 Std auf, was bei der beobachteten Mitosedauer von 1 Std einer Mitoserate von 0,033 pro Stunde entspricht. 1m gefarbten Knochenmark und Kulturausstrich wurden Zellen und Mitosephasen, auch unter Einwirkung von Spindelgiften, differenziert und die Ergebnisse statistisch ausgewertet. Der DNSStoff wechsel der Zellen wurde durch Mikroautoradiographien bestimmt.
