Metastasierender Krebs ist bei Erwachsenen in der Regel nicht heilbar. Eine Ausnahme stellen testikulre Keimzelltumoren (TKZT) dar, da ber 75 % der Patienten mit fortgeschrittenen metastasierenden TKZT mit einer auf Cisplatin basierenden Chemotherapie geheilt werden knnen. Ein besseres Verstndnis der molekularen Grundlagen der chemotherapeutischen Effizienz bei der Behandlung von Testistumoren knnte helfen, die Sensitivitt von anderen metastasierenden Tumoren zu erhhen. In dieser Arbeit wurde zum ersten Mal gezeigt, dass die Testistumorzellen in der Reparatur von InterstrangCrosslinks (ICL) defizient sind, wobei die verminderte ICLReparatur auf die niedrige Menge der DNAReparaturproteine ERCC1XPF zurckgefhrt werden konnte. Die Befunde der vorliegenden Studie zeigen, dass ERCC1XPF ein wichtiger Faktor ist, der die Resistenz gegenber Cisplatin beeinflussen kann. Ein gezieltes Targeting von ERCC1XPF knnte nun eingesetzt werden, um die ICLReparatur in Tumoren zu reduzieren und somit die Sensitivitt gegenber Cisplatin und weiteren ICLinduzierenden Agenzien zu erhhen. Somit knnte die Effizienz einer chemotherapeutischen Behandlung mglicherweise gesteigert werden.
