HlVspezifische CD8 TZellen spielen eine entscheidende Rolle bei der Kontrolle der Virmie in der akuten HIVInfektion. Im Verlauf der Infektion verlieren sie jedoch ihre Effektivitt. Die Ursachen hierfr sind bislang weitgehend unklar. In dieser Arbeit wurde zunchst die Expression des Zellaktivierungsmarkers CD38 und von PD1, einem Marker fr Immunerschpfung, auf HIVspezifischen CD8 TZellen untersucht. Die Ergebnisse zeigen, dass es bei fortschreitender Infektion zu einer Fehlregulation dieser beiden Marker kommt, was die Ineffektivitt der CD8 TZellen erklren knnte. Des weiteren wurde dem Ansatz nachgegangen, ob eine Kombination aus CD8 TZellantworten und einer antiretroviralen Therapie die Virmie zu kontrollieren vermag. CD8 TZellantworten gegenber allen wichtigen Resistenzmutationen konnten nachgewiesen werden, wobei der Focus auf die Lamivudinresistenz M184V gelegt wurde. Die Ergebnisse zeigen, dass eine Kontrolle der Virmie durch die Kombination einer antiretroviralen Therapie und CD8 TZellen mglich ist. Anwendung knnte dies in einer therapeutischen Vakzine finden.
