In den letzten Jahren, wurden immunsuppremierende Medikamente fr die Verbesserung des Kurzzeit Transplantationsergebnisses entwickelt. Das therapeutische Ziel besteht aus der Minimierung der Kosten bzw. der Minimierung der Nebenwirkungen verursacht durch die Immunsuppressiva und der Verbesserung des Langzeitresultates. Um eine Zerstrung des Transplantates durch die Immunsuppressiva zu vermeiden, bedarf es der Entwicklung von zustzlichen Toleranzprotokollen. Ein Kandidat fr die Toleranzinduktion in Transplantationen stellen die regulatorischen T Zellen dar. Da humane Tregs nur 12 % der peripheren CD4+ T Zellen reprsentieren, ist es nicht mglich ausreichende Zellmengen auf zu reinigen. Eine Etablierung adquater Protokolle fr die Isolation und Expansion ist notwendig. In unserem System ist es mglich Tregs mit einer hohen Reinheit zu isolieren und in verschiedenen Protokollen zu expandieren. Das suppressive Potential der expandierten Treg's wurde in drei funktionellen Assays bestimmt. Ein weiterer Aspekt dieses Projektes ist die Expansion isolierter Tregs mit Rapamycin
