Cette thse est consacre lapport de la modlisation 3D la neuroanatomie. Dans ce travail, nous nous intressons lorganisation spatiale de populations de neurones dont lapproche histologique classique tait dans lincapacit de fournir une vision comprhensive. Une premire partie montre que la modlisation 3D dvoile des agencements qui soustendent le mode de fonctionnement de groupes neuronaux et quelle a des vertus prdictives puisquelle ouvre de nouveaux questionnements par la mise en vidence dorganisations indites. Une deuxime partie rvle la difficult de comparer plusieurs modles 3D distincts et nous a conduits dvelopper un algorithme de moyennage et de fusion de modles ouvrant la porte la gnration de modles gnriques reprsentatifs. La dernire partie met en vidence que la fusion de modles 3D gnre des reconstructions biologiquement vraisemblables et permet dvaluer le degr de recouvrement de populations de neurones de plusieurs modles 3D. Ainsi par son approche quantitative, la modlisation 3D permet limage de saffranchir de son statut purement descriptif pour devenir, comme les autres paramtres biologiques, une grandeur quantifiable.
