Avant leur libration par exocytose, les neuromdiateurs sont accumuls dans les vsicules synaptiques par un transporteur vsiculaire dont la spcificit de substrat dtermine la nature du neuromdiateur libr. Des rgulations de leur activit pourraient aussi contrler la quantit du mdiateur vsiculaire. Pour aborder cette question, jai cherch caractriser les modifications posttraductionnelles de ces transporteurs. Un 1er travail a port sur les glycosylations du transporteur vsiculaire des monoamines (VMAT). Dune part jai confirm que VMAT porte bien 3 Nglycanes; dautre part jai dcouvert que VMAT porte aussi des Oglycanes dont la nature et le rle exacts restent lucider. Dans un 2nd travail, jai dcouvert lexistence dune phosphorylation du transporteur vsiculaire des acides amins inhibiteurs (VIAAT). Mes recherches sur sa signification biologique rvlent quelle ne rgule pas directement le remplissage des vsicules inhibitrices et quelle nest pas rgule par lactivit synaptique. En revanche, elle est induite au cours des tapes prcoces de la synaptogense, et pourrait donc tre associe une tape de la diffrenciation des neurones inhibiteurs.
