Les origines des troubles mnsiques non dmentiels observs au cours du vieillissement sont peu connus. Ces troubles pourraient rsulter de laltration primaire dautres fonctions neuropsychologiques, et en particulier, les troubles du sommeil, en raison de leur forte prvalence dans la population ge et de limplication du sommeil dans la consolidation de la mmoire. Les rsultats obtenus dans cette tude dmontrent que 33% 66% des troubles de la mmoire explicite lis lge sont explicables par laltration primaire du cycle veillesommeil. Nous montrons galement que ces altrations sont associes une fragmentation du sommeil lent et une dgnrescence des neurones cholinergiques du noyau pdonculopontin du tegmentum (PPT). Enfin, nous avons mis en vidence que deux mcanismes de rgulation du PPT, la voie de synthse des neurostrodes et la voie du transforming growth factor (TGF) sont galement altres.
