Protein biosynthesis is a core process in all livingorganisms. Assembly of the protein chain fromaminoacids is catalysed by the ribosome, an ancientand extremely complex macromolecular machine. Severaldifferent classes of accessory molecules are involvedin translation, and one set of them, calledtranslational GTPases (trGTPases) is the focus ofthis work. These proteins oscillate between theirGTP and GDPbound states via regulated cycles of GTPhydrolysis and exchange of GDP for GTP. The exchangeof GDP for GTP is accelerated by the GDP/GTP ExchangeFactor (GEF), while hydrolase activity of the GTPaseis stimulated by the GTPaseActivating Protein (GAP).The conformation of the GTPase in complex with GTPusually differs from that in complex with GDP, andthese structural changes serve as a signal for apartner molecule, inducing changes in its conformation.
